Sett's basic attacks alternate between left and right punch. Right punch is slightly stronger and faster. Sett also hates losing, gaining additional health regeneration based off of his missing health.

W (Haymaker) absorbs recent damage and sends it back: the center deals true damage. The more damage you take, the harder your W hits.
The trade combo is Q (speed + double empowered auto) into auto into auto into E (pull from both sides for stun) into W for the burst.
R (The Show Stopper) slams an enemy onto others — R the tank then slam them onto the carries for massive damage to the whole group.
The passive regenerates more when you're low on HP — Sett is dangerous even at low health.

How to counter and exploit weaknesses
Dodge the center of his W (true damage): sidestep laterally, the cone is predictable.
Don't deal too much damage at once without killing him: he stores the damage for an even stronger W.
Kite him: Sett has no dash (except targeted R) and relies on Q speed to gap-close.
In teamfights, don't group behind your tank: his R slams the tank onto the carries.
